Exploring the Role of FRAS1 Mutation in Medulloblastoma: Implications for Tumor Progression and Patient Survival

ABSTRACT 
Medulloblastoma is a malignant brain tumor common in children with high growth and metastasis potentials. This research aimed to investigate the role of the FRAS1 gene in medulloblastoma tumorigenesis and patient survival. Using cBioPortal, 791 medulloblastoma patient data were analyzed. Then, the impact of FRAS1 mutations on survival rates and potential involvement in tumor progression was examined. The siRNA-mediated FRAS1 knockdown in HTB-186 cell lines was performed to find the effect of siRNA-mediated FRAS1 knockdown on cell viability and gene expression. A significant correlation was found between FRAS1 mutations and decreased patient survival. This study found that the FRAS1 interactions with the GDNF pathway promoting cell survival were evidence of the FRAS1 involvement in tumor progression. In conclusion, the FRAS1 gene acts as an oncogenic driver in medulloblastoma. Therefore, targeting the FRAS1-GDNF pathway could offer new treatment options for medulloblastoma patients with FRAS1 mutations.​
